Sortilin inhibition by siRNA and the pharmacologic inhibitor AF38469 strongly reduced the adhesion and invasion of pancreatic cancer cells without affecting cell survival and viability. Sortilin inhibition also decreased the phosphorylation of the focal adhesion kinase in Tyr925. Together, these data show that sortilin contributes to pancreatic cancer invasion and could eventually be targeted in therapy.]]> Thu 24 Mar 2022 11:34:45 AEDT ]]> Nerve growth factor and its receptor tyrosine kinase TrkA are overexpressed in cervical squamous cell carcinoma https://nova.newcastle.edu.au/vital/access/ /manager/Repository/uon:41003 Thu 21 Jul 2022 10:51:13 AEST ]]> Proteomic analysis of neuroproteins in pancreatic cancer https://nova.newcastle.edu.au/vital/access/ /manager/Repository/uon:56016 Thu 18 Jul 2024 09:34:51 AEST ]]> The TrK receptor family https://nova.newcastle.edu.au/vital/access/ /manager/Repository/uon:26338 NTR, sortilin) that can interact with the Trks and affect their activity. The Trks are predominantly expressed in nervous tissues, where they are essential for the growth, development, and maintenance of select types of both peripheral and central neurons and are involved in neurodegenerative diseases. The Trks are also found in some nonneuronal tissues and in a wide variety of tumors, where they can play an active role in tumor progression and metastasis. The activated Trk receptors primarily bind and signal through Shc, FRS2, and PLCᵧ, which in turn activate PI3K, the Erks, and the hydrolysis of inositol phospholipids, among other events, leading to the modulation of gene transcription.]]> Sat 24 Mar 2018 07:35:55 AEDT ]]> Neurotrophin receptors TrkA, p75NTR, and sortilin are increased and targetable in thyroid cancer https://nova.newcastle.edu.au/vital/access/ /manager/Repository/uon:36867 NTR, and the proneurotrophin receptor sortilin were analyzed with immunohistochemistry in a cohort of thyroid cancers (n = 128) and compared with adenomas and normal thyroid tissues (n = 62). TrkA was detected in 20% of thyroid cancers, compared with none of the benign samples (P = 0.0007). TrkA expression was independent of histologic subtypes but associated with lymph node metastasis (P = 0.0148), suggesting the involvement of TrkA in tumor invasiveness. Nerves in the tumor microenvironment were positive for TrkA. p75NTR was overexpressed in anaplastic thyroid cancers compared with papillary and follicular subtypes (P < 0.0001). Sortilin was overexpressed in thyroid cancers compared with benign thyroid tissues (P < 0.0001). Neurotrophin receptor expression was confirmed in a panel of thyroid cancer cell lines at the mRNA and protein levels. Functional investigations using the anaplastic thyroid cancer cell line CAL-62 found that siRNA against TrkA, p75NTR, and sortilin decreased cell survival and cell migration through decreased SRC and ERK activation. Together, these data reveal TrkA, p75NTR, and sortilin as potential therapeutic targets in thyroid cancer.]]> Mon 12 Aug 2024 13:36:51 AEST ]]> Expression of NGF/proNGF and Their Receptors TrkA, p75NTR and Sortilin in Melanoma https://nova.newcastle.edu.au/vital/access/ /manager/Repository/uon:51398 Mon 04 Sep 2023 14:57:50 AEST ]]>
